PhybB_taRNA

taRNA under the control of the hybB promoter.

The hybB promotor is a coldshock promotor and therefore most activated at 30°C. The promotor is inactivated at a temperature of 37 °C. The concentration of hydrogen present in the cell can be a factor to consider looking at the influences of hydrogen concentration on the hybB promotor. (2)
In the AND gate of our system, repression of gene expression is achieved through sequestration of the RBS by formation of a stem loop structure by the 5'-untranslated region of the crRNA. This prevents binding of the RBS by the 30s ribosomal subunit and subsequent translation.
The taRNA in this part exhibits partial complementarity to the cis-repressive sequence. Its expression, which is driven by this hybB promoter, allows formation of a RNA duplex molecule and is accompanied by a conformational change in the crRNA structure. The stem-loop structure is unfolded, subsequently abolishing RBS obstruction and allowing translation of the target gene. (1)
