Applications of BBa_K103019

'Hunter and prey' selection system

This part was used as core of prey generator device. It was fused with:

protein A BBa_K103003. Resulting fusion was named His_alpha_A
protein Z_SPA-1 BBa_K103004. . Resulting fusion was named His_alpha_Z

Both fusions were overexpressed in BL21 strain. Due to ability of protein Z to form aggregates His_alpha_Z was purified simply by collecting post-sonication debris. His_alpha_A was purified on NiNTA-agarose bead (elution peak in 150 mM imidazole). The both prey protein fusions were added to culture media and used in selection experiments. Details here and here.
