Gold and QS ultra complex
The device is a tunable hybrid bistable switch. It receives two signals 1. gold (HAuCl4) and 2. population density, which modify magnitude of the output 'transactivation of the target gene'.
LuxR [1] is produced constitutively by the promoter BBa_J23100 [2]. It is activated by the quorum sensing signal AHL and forms a positive feedback loop to promote expression of the AHL synthase LuxI [3] and the activator taR12 [4]. The latter binds to crR12 [5] preceding translation start site of the mRNA of the target gene resulting in the full expression. Gold is recognized by the part BBa_K1127008 [6] which in response produces AiiA [7]. AiiA can degrade AHL; therefore, it reduces expression of the target gene.
