This origin propagates at the permissive temperature of 30&deg;C.  At 37&deg;C, the restrictive temperature, the repA protein becomes non-functional so that plasmids containing this origin can no longer propagate.  It will take a few cell divisions for plasmid-free cells to arise and overtake the population.

BBa_I50052 is a pSC101 temperature sensitive origin of replication. BBa_I50052 is the temperature sensitive replication origin found in the pSB8* series of BioBrick vectors as well as the pSIM5 recombineering vector.

Usage and Biology

This origin propagates at the permissive temperature of 30°C. At 37°C, the restrictive temperature, the repA protein becomes non-functional so that plasmids containing this origin can no longer propagate. It will take a few cell divisions for plasmid-free cells to arise and overtake the population.
