AND gate
The AND gate is a type of Boolean logic gate that responds to the corresponding upstream promoter only if all markers are present, thus activating downstream gene expression.
Usage and Biology
The AND gate is constructed using the p7 promoter and two terminators ( BBa_B0015 , BBa_J61048 ). On both sides of the terminator BBa_B0015, we introduced the cis-Tp901 integrase site attB TP901 and the trans-Tp901 integrase site attP TP901. Similarly, on both sides of the terminator BBa_J61048, we introduced the cis-Bxb1 integrase site attB Bxb1 and the trans-Bxb1 integrase site attP Bxb1 to form the so-called AND gate. to constitute the so-called AND gate. Finally, we introduced the RiboJ and BCD2 sequences to ensure smooth transcription with the OR gate on.
The AND gate can be assembled to any biobrick containing an RBS site. The AND gate can be used to respond to multiple promoters upstream, but only if all markers are present. To demonstrate its efficacy, we designed the RFP protein downstream of it to test it ( BBa_K4156116 ).
Meanwhile, we plan to apply the AND gate on the expression of HlyE ( BBa_K4156120 ). Together with the use of upstream regulatory sequences, we can control the expression of HlyE only when needed, while avoiding any undesired production and harm.
