TetR represses the PtetO promoter, producing NarP. In the presence of both nitrate and nitrite, NarQ phosphorylates NarP. Therefore, phosphorylated NarP measures both nitrite and nitrate levels. By using the corrected difference, our NarP and NarL (Lambert iGEM’s improved part of HKUST-Rice 2015 <a href=“https://parts.igem.org/Part:BBa_K1682018”>BBa_K3411020</a>) composite parts are able to evaluate the nitrite levels.

Description

The listed NarL’s role within Lambert_GA iGEM 2020’s (improved from HKUST-Rice 2015, BBa_K1682018) system serves as an acceptor of the phosphate group from NarX. The promoter BBa_J23106, a constitutive Anderson Promoter, initiates the production of TetR. TetR then represses the PtetO promoter. By regulating the amount of aTc, the molecule that inhibits TetR, the team can then control levels of NarL. In the presence of nitrate, NarX phosphorylates NarL. Then, phosphorylated NarL binds to a specified promoter and correlates to measured nitrate levels.
