RNA Part of an RNA·DNA-DNA triple helix + MS2-Stemloop
This part consists of a sequence that forms an RNA·DNA-DNA triple helix (DNA part: BBa_K3657023) and a sequence that forms an MS2 hairpin (BBa_K3113006).
Usage and Biology
The MS2 hairpin binds to the MS2 protein (BBa_K3218006). The triple helix forming part binds the DNA sequence specifically. In that way this part should theoretically attract a protein of interest fused to an MS2 protein to a specific region on DNA. Due to the COVID19 pandemic we were not able to get any empirical evidence that this part functions as described above. More information available here: iGEM Heidelberg 2020.
Figure 1: Secondary structure of BBa_K3657025
MS2 hairpin forms a stable structure. The triple helix forming sequence doesn't form any stable structure and does not interfere with the MS2 hairpin. The structure was generated by the Geneious software.
