SV40 promoter is a viral promoter derived from Simian Virus 40.
Because we could not find the SV40 promotor in the parts registry, the Freiburg iGEM Team 2013 decided to provide one to the following teams. This enables the teams to choose from differently strong mammalian promoters – dependent of the individual intention. We tested our own devices with both promoters (BBa_K1150011 [SV40], BBa_K747096 [CMV]). For further information please take a look at BBa_K1150019 and BBa_K1150020 or click [http://2013.igem.org/Team:Freiburg/Project/effector#activation here].
In an earlier study CMV and SV40 promoter were tested in different B and T cells as well as in two non-lymphoid cells. In all cell lines the CMV promoter construct was higher expressed than that of SV40 [Zarrin et al., 1999].
In comparison to the cellular promoter VV1 SV40 showed similar activities. Hence with an SV40 promoter one can gain a more physiologic control for reporter assays compared to CMV.
