Edinburgh can confirm that the sequence of the part itself is as expected. The part comes in RFC12 format and therefore has an unusual prefix and suffix. We found an extra C base in the suffix at the end of the NotI site: Expected: GCTAGC GCGGCCG CTGCAG Actual: GCTAGC GCGGCCGC CTGCAG Edinburgh successfully fused this part to Enhanced Yellow Fluorescent Protein (with a short linker sequence) to produce BBa_K523013. Since the cells fluoresced, the INP (plus linker) did not prevent correct folding. Furthermore, we found evidence that YFP was localised to the cell membrane. |
