This contribution adds useful information to understand how the signal peptide works.

The SEC-N22 signal peptide originates from the curli secretion pathway (Type VIII) (1). This pathway is natively used in E. coli to secrete monomeric protein units of curli fibres (CsgA) which self-assemble into membrane-bound extracellular curli fibres during biofilm formation (2,3). This pathway is composed of seven curli-specific genes (csg) encoded by two operons (csgBAC and csgDEFG). These operons encode the structural subunit of the curli protein and other accessory proteins required for CsgA secretion and assembly (4). CsgA is secreted into the periplasm with the help of the SEC (1-60) peptide and across the outer membrane with the help of the N22 peptide(61-126) (4). These two components are essential for the secretion of proteins through this pathway and are not specific to CsgA. SEC-N22 can thus be fused at the N-termini of your gene of interest for the protein to be secreted extracellularly.
