Pirin like protein (pirA) from Synechocystis sp PCC6803

Protein that is supposed to increase pyruvate levels in Synechocystis sp PCC6803 in concert with pirB. (BBa_K273003).

Mechanism unknown, function yet to proof.


Usage and Biology

The protein Pirin has in Serratia marcescens been shown to regulate the pyruvate levels by means of inhibition of the PDC (Po-Chi Soo et al. JOURNAL OF BACTERIOLOGY, Jan. 2007, p. 109–118). While the Pirin superfamily is diverse and has different functions across organisms it has been linked to growth inhibition in Synechocystis sp. PCC 6803 (Hihara et al. FEBS Letters 574 (2004) 101–105). One could theorize that it does this in the same way as in S.marcescens, by inhibiting the PDC and thus choke the krebs cycle. In S. marcescens this leads to induction of fermentation pathways.

In Synecocystis two pirin class proteins, pirA and pirB seems to act in concert (Hihara et al. FEBS Letters 574 (2004) 101–105).

The Uppsala iGEM team of 2009 plan to test these proteins in order to increase substrate levels for ethanol synthesis.
