RFC12 Internal Chitin Binding Domain

This is a RFC12 (SpeI,NheI) compatible chitin binding domain. It is an internal domain meant for either N or C terminal fusions.

Usage and Biology

Chitin binding domains are most commonly used with self-splicing intein tags such as BBa_J70628. CBD tags are used primarily for protein purification due to the high yields and purity, and along with an intein domain the tag can be easily removed. Note that the N or C terminal residue of the tagged protein can affect intein cleavage. See BBa_J70629 for the intein-CBD fusion.

Common tag information:

  • Matrix: Chitin
  • Elution: DTT or B-ME
  • Yield/Purity: low milligram yields, 100% purity
  • # Steps: One step
  • Fusion Location: Internal
  • Affects Function?: Possibly, unlikely after intein cleavage
