ComP

Usage and Biology

ComP is a transmembrane protein from B. subtilis which is a quorum-pheromone receptor. Upon binding to the gram-positive quorum molecule ComX (a secreted 10-residue modified peptide), ComP will phosphorylate the downstream response regulator, ComA. The membrane topology of ComP has been investigated, but the crystal/NMR structure is unknown.

Technical Notes

1. The ComP gene was isolated by PCR from genomic DNA of B.subtilis 168.

2. Due to a large percentage of A+Ts, the primer hybridization sequence was lengthened and the Biobrick sites were correspondingly shortened. Thus, the part does not contain the canonical Biobrick sites (EcoRI-NotI-XbaI, SpeI-NotI-PstI), but are instead shorter BB sites (EcoRI-XbaI, SpeI-PstI).

3. This part contains an SpeI site, which will be removed by site-directed mutagenesis in the future.
