NYU Abu Dhabi 2024 Characterization

NYU-Abu-Dhabi 2024 BBa_B0030_m0 was assembled into a construct using Golden Gate Assembly, with eYFP as a reporter to measure its ribosome binding site (RBS) activity via fluorescence at two temperatures: 28°C and 37°C. According to the RBS activity graph, BBa_B0030_m0 (represented by the red lines) showed a distinct pattern at both temperatures. At 37°C (solid red line), the RBS displayed low initial activity followed by a steady increase, peaking before gradually declining toward the end of the experiment. At 28°C (dashed red line), BBa_B0030_m0 showed a similar trend, with an initial increase in activity that peaked earlier, followed by a gradual decline. However, the overall activity at 28°C was slightly lower compared to 37°C. These results indicate that BBa_B0030_m0 shows a peak-and-decline pattern at both temperatures.
