2,3-DHB Biosynthesis Construct, with entC and ahpC

This device codes for enzymes that are part of the biosynthetic pathway of enterobactin, a compound with a strong affinity for iron (III) ions, which are naturally produced by bacteria in order to sequester this essential trace nutrient. The specific aim of this construct is increasing the production of 2,3-dihydroxybenzoic acid (2,3-DHB), an intermediate that is itself a strong coordinator to Fe (III) ions; upregulating production of this small molecule should grant transformed bacteria the ability to increase dissolution of less soluble compounds like iron (III) oxide. During its biosynthesis, entC (isochorismate synthetase) and ahpC (alkyl hydroperoxide reductase) work together to catalyze the critical transformation of chorismate to isochorismate, which is subsequently converted to the target by other cytosolic enzymes. The coding sequences were obtained from reverse translation and optimization of UniProt amino acid sequences, and other parts were obtained from the iGEM parts registry.
