Design Notes

We needed a way to make sure that E.coli placed in the lungs of CF patients will die upon leaving that environment. To this end, we decided on using an AND gate structure to make sure we have strict regulation needing two signals. One signal is the Sialic acid. This is found in mucosal areas which include the lungs. To narrow down the areas which we want, we added the necessity for another signal, LasL. This is a quorum sensing molecule used by the bacteria causing the infection. With the necessity for these two signals, we greatly narrow the conditions of where this E.coli can survive, leaving only really the lung environment for CF patients.

The mechanism of death is in the absence of any of the two signals, Granulysin and DNase1 will be produce which will kill the cell.
