Unfortunately, no data was collected pertaining to the functionality of this part and further experiments must be conducted to determine its performance. Upon transformation, we found that our plates were consistently contaminated and only bacterial colonies grew. While the complete contamination made us unable to asses the functionality of the part, it may also indicate something about the part; perhaps this part does not make for very viable yeast colonies. If this is the case, this could be due to the fact that we put the gene into a vector that contained a constitutive promoter. Therefore, once the cells were transformed they began to dedicate too many of their resources to expressing LacI protein, thus inhibiting the cells from growing up properly.
Applications of BBa_M36525
This part, in conjunction with a promoter containing a LacI operon binding site, can be used to make an IPTG or Lactose induced logic gate in yeast. This has potential for a lactose sensor (which would be beneficial for lactose tolerant individuals) or simply in creating a more complex logic gate or circuit.
