PAmCherry (photoactivatable red fluorescent protein)
PAmCherry is a photoactivatable red fluorescent protein. PAmCherry fluoresces when exposed to 350–400 nm light while does not fluoresce at other wavelengths. It is used a reporter tag; by expressing in cells and localizing it to sub-cellular locations, one can gain spatial information within the cell by tracking organelles and proteins. The photoactivated PAmCherry can be visualized with the same filter sets used for other red fluorescent proteins.
Both images are of PAmCherry expressed in the yeast cell targeted to the cytosol. The left image shows PAmCherry prior to being photoactivated. There is a diffuse low level of fluorescence before activation. The right image was taken post-activation of PAmCherry showing fluorescence magnitudes brighter.
