GAL4 DNA binding domain
This part contains the DNA binding domain of the transcription activator GAL4 (GAL4DBD). This part is flanked by RFC25 pre- and suffix to allow N- and C-terminal protein fusions.
If this part is used in yeast and you want to fuse your protein to the N-terminus of this part, it might be nescessary to add a nuclear localization sequence (NLS) (BBa_K801030, SV40 NLS) to the N- or C-termini of the fusion protein because the endogenous N-terminal nuclear localization signal of GAL4DBD can be masked by the fused protein. If you only want to do C-terminal fusion to this part, the endogenous NLS is sufficient to translocate your whole fusion protein into the nucleus.
Additionally, one can add a protein linker between two protein domains to improve independent folding of the domains. If you want to fuse one protein domain to the N-terminus of GAL4DBD and also want to use a linker in between, there is already a composite part (BBa_K801037) of a 20 amino acids linker fused to the N-terminus of this part (N'-20aaLinker-GAL4DBD-C').
