Design Notes
When amplifying this part, the overlapping regions were carefully chosen so that the insert sequence alone can be inforporated into a plasmid, for future use in overlap extension. Digesting with the two inner enzymes for the BB-2 enzymes S and N, leaves the appropriate sequence for overlap extension.
Source
The original XylE came from a biobrick part. It was amplified using PCR and then overlap extension was used to incorporate it into the flagella.
