rrnbT1 rrnbT2 terminator

Usage:

In the Bac'Attack Aix-Marseille University team's project, the rrnB T1 T2 terminator is employed to ensure efficient transcriptional termination and prevent read-through transcription. Originating from E. coli and derived from the plasmid Pbad-HisC, this strong bidirectional terminator sequence is placed downstream of the coding regions of all the composite parts of the project.

Biology:

The rrnB T1 T2 terminator consists of two strong transcriptional termination sequences (T1 and T2) sourced from the E. coli ribosomal RNA operon rrnB. Terminators are critical regulatory elements in prokaryotic gene expression, ensuring that transcription stops efficiently once an mRNA transcript is complete. This prevents the RNA polymerase from continuing past the intended gene, which could lead to unwanted or uncontrolled expression of downstream genes.

The T1 and T2 terminators function by inducing the formation of a stable stem-loop structure in the nascent mRNA, causing RNA polymerase to dissociate from the DNA template, thereby halting transcription. This mechanism prevents transcriptional read-through, which is particularly important in polycistronic systems or when multiple constructs are present in the same operon. The use of strong terminators like rrnB T1 T2 enhances the stability of mRNA and improves the efficiency of protein production, making it a valuable tool in synthetic biology.
