Design Notes
Histidine tag, also known as His-tag, is one of the simplest and most widely used purification tags with six or more consecutive histidine residues, which can be inserted into the C-terminus or N-terminus of the target protein.
Source
Currently, the purification of genetically engineered proteins with His-tag is mostly performed by immobilized metal chelate chromatography, which utilizes the interaction between the transition state metal ions and the histidine side chain to isolate the target protein.By literature:LIU Chengjun, HUANG Qing, ZHAO Lian, et al.
