Fine control device
We designed a fine tuning device using paT7P-1 (part: BBa K5328009- parts. igem. org), G8 Gluc (part: BBa K5328010- parts. igem. org), and T7 promoter (part: BBa K5328015- parts. igem. org). We added the T7 promoter before the target gene that needs to be expressed, so that RNA polymerase in normal bacteria cannot initiate its expression. We will secrete G8 Gluc into the bacterial environment for specific detection and binding to AFB1. After that, Gluc will restore its activity, and oxidized coelicin will emit blue fluorescence. Blue fluorescence enters the cell as a signaling molecule. After receiving the signal, the VVD domain undergoes conformational changes, causing the protein expressed in paT7P-1 to form a homodimer. The cleaved T7 RNA polymerase regains activity, recognizes and binds to the T7 promoter, and initiates the expression of the target gene. This device achieves precise automatic regulation of the expression of a certain substance. We also provide various nanobodies that can identify different substances.
