The target site of mutation is chosen referring to the domain function analysis in the article "Hydrolytic endonucleolytic ribozyme (HYER)is programmable for sequence-specific DNA cleavage", which explains the function of the 2nt-bulge as the structure which DNA substrate stabilizer to help with locating.
