Autotransporter
Signal peptides (SPs) are short amino acid sequences that control protein secretion and translocation in all living organisms. And the amino acid sequence of EstA indicated that the esterase is a member of a novel GDSL family of lipolytic enzymes. The estA gene showed high similarity to an open reading frame of unknown function located in the trpE-trpG region of P. putida and to a gene encoding an outer membrane esterase of Salmonella typhimurium. Both of them consist of the autotransporter.
Usage and Biology
The autotransporter is used to transport the target protein to the cell membrane based on its characteristic of cell menbrane localization.
Fig.1 The schema of the autotransporter's mechanism.
In our project, we use it to transport PEBP-PEase onto the cell membrane to bind to the PE particles as well as degrading them into monomers.
