Design Notes
During the design of this part, special attention was given to the accurate representation of the seven-transmembrane domain structure critical for GPCR function. The receptor's ability to couple with Gi/o proteins and its structural features, such as the ECL2 "lid-like" structure, were key considerations to ensure proper ligand binding and signaling function. Additionally, potential challenges such as membrane insertion and expression levels in mammalian cells were considered to optimize the receptor's functionality in synthetic biology applications.
Source
The MT1 receptor sequence is derived from mammalian species, primarily based on the human MT1 receptor gene. The receptor is a well-characterized member of the GPCR family and is widely studied for its role in circadian rhythm regulation.
