The zwf gene in Escherichia coli encodes glucose-6-phosphate dehydrogenase (G6PDH), a key metabolic enzyme that plays a role in the pentose phosphate pathway (PPP), also known as the hexose monophosphate shunt or hexose phosphate pathway. The reaction catalyzed by G6PDH is the conversion of glucose-6-phosphate (G6P) to 6-phosphogluconolactone, while producing NADPH. NADPH is an important reducing power carrier within cells, used in various synthetic metabolic processes, including fatty acid synthesis, cholesterol synthesis, and defense against oxidative stress. In synthetic biology and metabolic engineering, enhancing the expression of the zwf gene can increase PPP flux, thereby increasing the supply of NADPH, which is crucial for increasing the yield of certain metabolic products within cells.
