We found an optogenetic tool for far-red light-regulated transcriptional activation apparatus that has been well characterised. During daytime,far-red light initiates the production of MrkH in our circuit.PMrkA is an inducer promoter that recognizes and binds to MrkH.Under the activation of PMrkA, downstream gene Cl are expressed. It encodes the Cl repressor protein which binds to Plam and inhibits the expression of downstream LuxCDEFG.As a result, there are no products of LuxC, LuxD and LuxE to synthesise the substrate for the light emitting reaction.This largely saves the energy and substrates for luminescence at night.
Usage and Biology
In the light Emitting Module, sunlight activation is limited by the UV-B content and it fails to activatie the promoter PumuDC and the downstream LuxAB. So we found an optogenetic tool for far-red light-regulated transcriptional activation apparatus that has been well characterised.
PMrkA is an inducer promoter that recognizes and binds to MrkH, and recruits RNA polymerase by transcription factor MrkH to activate the expression of downstream genes. MRKH recruits RNA polymerase and specifically binds promoter PMrkA, activating transcriptional expression of LuxAB.
