ScrR

ScrR plays an important role in regulating sucrose metabolism[1].The nucleotide sequence of the regulatory gene scrR comprises an open reading frame that specifies a protein of 35.8 kDa. This protein exhibits similarity to transcriptional regulators of the GalR-LacI family[2].It exerts its negative effect on transcription initiation.

Usage and Biology

In the cycle of preventing false positives and leaks, in order to improve the sensing of sucrose ,we design Line II. Line II enabled the engineered bacteria to activate the sensing of sucrose only in the hypertonic environment of the ocean, and after entering the ocean, the spare sucrose in the product and the sucrose produced by the algae inhibited the deterrence of Pscr by ScrR, which led to the expression of lacI and the non-expression of MazF. After the engineered bacteria were detached from the system, the sucrose concentration was reduced, the inhibition of Pscr by ScrR was enhanced, the expression of lacI was reduced, and MazF was then expressed to play a killing role. We use this part for the composite part BBa_K5210019.

This test basically verified the inhibitory effect of sucrose on ScrR, and it was also an exploratory test.
