promoter umuDC (UV promoter,Puv)

The umuDC promoter is a key regulatory element in the SOS response of E. coli, which is located upstream of the umuD and umuC genes and controls the expression of these genes under conditions of DNA damage stress[1]. umuDC promoter expression is repressed by LexA proteins, but upon RecA activation, LexA is degraded, which initiates the transcription of umuDC. The proteins encoded by the umuD and umuC genes regulated by this promoter are essential for UV and chemically induced mutagenesis and are involved in the translesion synthesis (TLS) mechanism of damaged DNA[2].


Usage and Biology

In our genetic wiring, promoter umuDC is controlled by UV light, which senses changes in UV intensity. When UV irradiated ssRNA accumulates and binds to RecA causing it to be unable to cleave LexA. the inhibitory effect of LexA on promoter umuDC is canceled, initiating downstream gene expression.

Experimental results

We introduced the plasmid pet28a-PumuDC-EGFP into E. coli. The transformed E. coli was irradiated using a UV lamp. Compare the expression of reporter gene EGFP after treatment with different irradiation time.

We did not observe the activity of this promoter well in the laboratory due to the overpowering bacterial killing effect of UV lamps.
