As part of our project we designed a number of iterations on the P4 cosmid, of which the RFP cosmid is only one. Producing transducing units with each of those devices yielded different titers of transducing units depending on the total size of the device and plasmid backbone. We have found that packing of a plasmid which includes one of these devices will occur at highest efficiency when the total size of plasmid is close to that of wild type P4 (~11.6kb). Reducing the size of the plasmid to ~7kb results in titers several orders of magnitude smaller than when produced with the P4 cosmid, but can still produce viable transducing units.
