B-Arrestin-2-TEV-Protease MammoBlock

This part encodes a fusion between the N1a catalytic proteolytic domain of the tobacco etch virus (TEV) and the human B-Arrestin-2 sequence. The N1a domain recognizes the amino acid sequence ENLYFQX (where X is a variable amino acid) with unparalleled specificity, and is typically used for the removal of tags made for protein purification. The B-Arrestin-2 protein induces the extinction of nonvisual G-coupled protein receptor (GPCR) signaling in mammalian cells by binding to the cytoplasmic tails of induced GPCRs. This part is capable of cleaving GPCR-TEVs-Transactivator fusion proteins and freeing the tethered transactivator to induce transcriptional changes in a cell's nucleus.
