It is located downstream the coding sequence of an enzyme in an expression cassette, dehalogenase type II in part K5109001. It should be used for golden gate assembly only with elements that are designed with specific compatible ends leaved after BsaI cleaving. In this case, the sequence was designed to allow insertion of terminators from the 2024 distribution kit, that present compatible ends after cleavage with BsaI.
The part solves it's purpose when considered inside the complete context of use, in this case, part K5109901. In order to perform Golden Gate cloning on it, it should be located either into a plasmid, or into a sequence of interest.
