T7 promoter, The downstream T7 promoter incorporated one DNA sequence, which encoded a 6-his tag, to bind to the nickel-resin column for the in vitro protein purification. T7 promoter can be manipulated in the presence of isopropyl β-Dthiogalactoside (IPTG) to trigger our composition parts generating more RNA, and later on for the abundance protein expression to extract our team’s cloned enzyme(s) via the in vitro protein purification as the future experiments. However, our team will still express the mutants along with the wild-type created, respectively, in vivo for the experiments.
