The hsa-mir-99b_2DDSD_s (BBa_K5086109) is formed by the combination of hsa-mir-99b_2DDSD (BBa_K5086019), hsa-mir-99b_target (BBa_K5086000), and two wt (BBa_K5086017) chains. Among them, the sequence at the 5' end of hsa-mir-99b_2DDSD is successively connected to the hsa-mir-99b_target and the two wt chains in a complementary pair, and the sequence at the 3' end of it is complementary to the chain of Universalness_h (BBa_K5086009), so that the hsa- mir-99b_2DDSD_s can bind specifically to the Universalness_h strand thus accomplishing the recognition of the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction starting from the 3' end of the target hsa-mir-99b_Rep. During DNA polymerization by Bst DNA polymerase, the wt strand is replaced and released by the newly synthesized daughter strand, which also releases a DNA single strand with the same sequence as the target At the same time, a single strand of DNA with the same sequence as the target strand (U to T) will be released, which will serve as the target for the first step of the polymerase-mediated strand displacement reaction to realize the DDSD self-enhancement process, and the wt strand will be involved in the subsequent reaction to produce a fluorescent report.
