Universalness_RP1 (BBa_K5086108) is formed by complementary pairwise ligation of Universalness_F (BBa_K5086014) and Universalness_Q (BBa_K5086016), due to the modification of quenching motif BHQ1 at the 5' end of Universalness_Q, while the 3' end of Universalness_F is modified with fluorescent motif FAM. Q (BBa_K5086014) and Universalness_Q (BBa_K5086016) are complementarily paired, due to the modification of the quenching motif BHQ1 at the 5' end of Universalness_Q, and the modification of the fluorescent motif FAM at the 3' end of Universalness_F, which together form a group of reporter probes called Universalness_RP1. When the target nucleic acid is not present, the reporter probes are quenched due to the formation of a stable double-stranded structure, which closes the distance between the fluorescent molecules and the quenching molecules, resulting in fluorescence resonance energy transfer (FRET), and fluorescence quenching; when the target is added, the wt strand of polymerase-mediated strand displacement hybridizes with the long DNA strand in the double-stranded probes through the toehold-mediated strand displacement reaction to form a thermodynamically more When the target is added, the wt chain from the polymerase-mediated chain replacement hybridizes with the DNA long chain in the double-stranded probe through the toe-mediated chain replacement reaction, resulting in the formation of a thermodynamically more stable double-stranded structure, and at the same time competes with the DNA short chain for fluorescence recovery, releasing fluorescence signal.
