The hsa-mir-21_Rep_s (BBa_K5086102) is a composite element formed by combining hsa-mir-21_Rep (BBa_K5086007) and Universalness_h (BBa_K5086009). The 5' end of hsa-mir-21_Rep is complementarily paired with the Universalness_h chain, and the remaining sequence at the 3' end of hsa-mir-21_Rep in hsa-mir-21_Rep_s is complementary to the target hsa-mir-21, thus it can The remaining sequence at the 3' end of hsa-mir-21_Rep_s is complementary to the target hsa-mir-21, so it can specifically bind to the target to recognize the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction from the 3' end of the target hsa-mir-21, and during the DNA polymerization process by Bst DNA polymerase, the Universalness_h strand is replaced by the newly synthesized daughter strand and released. Through this process, the target signal is converted to the Universalness_h strand, which will continue to undergo subsequent strand replacement reactions to produce a fluorescent report. (The significance of this chain replacement step is that it facilitates the subsequent self-enhancement, and the DDSD probe shelf cannot be synthesized without the replacement of the Universalness_h chain.)
