hsa-mir-200c_Rep_s (BBa_K5086101) is a composite element formed by the combination of hsa-mir-200c_Rep (BBa_K5086006) and Universalness_h (BBa_K5086009). In which the 5' end of hsa-mir-200c_Rep is connected by complementary pairing with the Universalness_h chain, and the remaining sequence of the 3' end of hsa-mir-200c_Rep_s in hsa-mir-200c_Rep_s is able to be connected to the target hsa-mir- 200c, so it can bind specifically to the target to recognize the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction from the 3' end of the target hsa-mir-200c, and during DNA polymerization by Bst DNA polymerase, the Universalness_h strand is replaced and released by the newly synthesized daughter strand. Through this process, the target signal is converted to the Universalness_h strand, which continues to undergo subsequent strand replacement reactions to produce a fluorescent report. (The significance of this chain replacement step is that it facilitates the subsequent self-enhancement, and the DDSD probe shelf cannot be synthesized without the replacement of the Universalness_h chain.)
