hsa-mir-141_2DDSD (BBa_K5086022) is a single-stranded DNA sequence with 88 nucleotides that is a self-enhancing probe for the target hsa-mir-141_Rep. The sequence at its 3' end is designed to be complementary to the Universalness_h strand so that it binds specifically to the Universalness_h strand and thus accomplishes recognition of the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction from the 3' end of the target hsa-mir-141_Rep. During DNA polymerization by Bst DNA polymerase, the wt strand is replaced and released by the newly synthesized daughter strand, and the DNA single strand whose sequence is identical to that of the target strand is released as well. At the same time, a single strand of DNA with the same sequence as the target strand (U to T) will be released, which will serve as the target for the first step of the polymerase-mediated strand displacement reaction to realize the DDSD self-enhancement process, and the wt strand will be involved in the subsequent reaction to produce the fluorescence report.
