Universalness_RP2 (BBa_K5086015) is a single-stranded DNA sequence with 18 nucleotides and modified with the fluorescent motif BHQ at the 3' end and the quenching motif at the 5' end. In the absence of the target nucleic acid, the proximity between the fluorescent molecules and the quenching molecules limited by the DNA strand results in fluorescence resonance energy transfer (FRET) occurring and fluorescence quenching; when the target is added, the wt strand mediated by polymerase-mediated strand displacement activates the paracrine activity of cas12A enzyme in conjunction with the Universalness_crRNA to perform non-specific cleavage to cut the strand into two segments, achieving fluorescence recovery and releasing a fluorescent signal.
