hsa-mir-141_Rep (BBa_K5086008) is a 52-nucleotide single-stranded DNA sequence that is a strand displacement probe for the target hsa-mir-141. Its 5' end is designed to be complementary to the Universalness_h strand, and the sequence at the 3' end is designed to be complementary to the target hsa-mir-141, so that it binds specifically to the target and thus accomplishes recognition of the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction starting from the 3' end of the target hsa-mir-141. During DNA polymerization by Bst DNA polymerase, the Universalness_h strand is displaced and released by the newly synthesized daughter strand. Through this process, the target signal is converted to the Universalness_h strand, which will continue to undergo subsequent strand replacement reactions to produce a fluorescent report. (The significance of this chain replacement step is that it facilitates the subsequent self-enhancement, and the DDSD probe shelf cannot be synthesized without the replacement of the Universalness_h chain.)
