Resilin Oligo SacI 2 end

The aim was to generate 64 repeats of the resilin sequence (BBa_K5077000). In order to achieve this, the Repeatigo method was developed. Six distinct Oligonucleotides are necessary for this method. This specific Oligonucleotide is one of them. Further details can be found in the description of part BBa_K5077019.

What´s so special about this part?

This part is complementary to Oligo SacI 1 end (BBa_K5077005) and creates the end sequence of the desired sequence. After these two bind to the middle oligos (Oligo 1 repeat and Oligo 2 repeat) that created the main middle part, they cannot bind any more oligos because Oligo SacI 1 end and Oligo SacI 2 end will create blunt ends. The aligned end oligos will add the restriction site to the sequence for SacI, which is needed to clone the sequence into pET28c(+) for expression in E. coli.
