Resilin Oligo 2 repeat

The aim was to generate 64 repeats of the resilin sequence (BBa_K5077000). In order to achieve this, the Repeatigo method was developed. Six distinct Oligonucleotides are necessary for this method. This specific Oligonucleotide is one of them. Further details can be found in the description of part BBa_K5077019.

What´s so special about this part?

This oligo represents the resilin exon as a whole, but the exon is arranged differently. It is made up of complementary bases and is shifted by half. This means that when this oligo binds to the oligo 1 repeat BBa_K5077001), they will align for 23 bp even though they are 45 bp long. This creates overhangs on either side and this middle part of the desired sequence can grow on either side.
