In order to inhibit the infection of the Beet Yellows Virus in sugar beets, we designed lhRNAs targeting the virus genome. One of the target sequences chosen for our lhRNAs is the one coding for the RNA-dependent RNA polymerase (RdRp). The sequence coding for the RdRp is highly conserved in the virus, meaning that it is not truly mutating with time, and is responsible for synthesizing viral progeny in infected cells. Inhibiting it will result in a lack of virus replication, and then its elimination. We kept a sequence of 569 bases (RNA-directed RNA polymerase homolog - sugar beet yellows virus (fragment) [Beet yellows virus] on NCBI). This part encodes the lhRNA sequence against the RdRp of the BYV that will then be inserted in an expression plasmid to produce the lhRNAs.
