Design Notes

Throughout the light induction experiment, the ASR is inserted in the pET-17b-ASR, and the Flag-tag is fused with ASR protein as the ASR inhibits the expression of genes in the dark. In specific, when the C-terminus of ASR binds to the DNA, in this case, is the BBa_K2596001 above, ASR will inhibit gene expression.
