Usage and Biology
LIRA_input05 is the target to be detected by LIRA_H05, it will bind to the loop at the top of LIRA_H05, and then change its conformation so that the ribosome could bind to the RBS.

This composite part is used to express input05 in E.coli to test the function of H05.
Characterization
T7 promoter could be induced by IPTG, so the intracellularly concentration of LIRA_input05 produces a large difference between the two conditions of IPTG presence/absence.
Compared to BBa_K5038024, this plasmid has an additional lac operator downstream of T7 promoter, which is the binding site of LacI repression protein and could further prevent the transcription leakage.
