Usage and Biology
This linker is used to connect LIRA hairpin and the reporter gene. In our experiments, we use EGFP, lacZ, or lacZα as the reporter gene. The presence of linker makes the LIRA system more stable i.e., more conducive to expressing reporter gene and indicate the existence of target RNA.

Characterization
We simulated the RNA secondary structure of the LIRA-reporter system by NUPACK, using the presence or absence of the linker as a control, and assessed the stability of the RNA secondary structure by its free energy, the higher the free energy, the lower the stability. The data are summarized in Figure 2.

Temperature: 37℃, Unit: kcal/mol
As shown in Figure 2, in most LIRA-reporter systems, the free energy in the absence of a linker is greater than that in the presence of a linker, which indicates that the linker helps enhance the stability of the LIRA-reporter system.
