Design Notes
The initial version of our SRE promoter had repeat structures that caused synthesis problems. We modified the design to reduce the repeat homology at nonconserved positions and have since verified that this did not disrupt promoter activity. We know it is synthesizable since we already had it synthesized and are testing it in a luciferase-based system. However, we believe the BFP/YFP Asimov system will provide better internal controls and provide a more robust and less noisy reporting system.

Our SRE promoter is based on the strong constitutive CMV promoters, which is available as an open source part (BBa_I712004). Our modifications, the insert of the SRE element, were our own work and thus can be accessed as open source once we release it.
