Design Notes
1 Due to the polyT structure being a termination signal for RNA polymerase III, it is necessary to avoid having four or more consecutive T's in the shRNA sequence. 2 Avoid target sites that overlap with single nucleotide polymorphism (SNP) regions. 3 The U6 promoter can only transcribe relatively small gene sequences (mostly gene fragments less than 100bp), and cannot transcribe protein-coding genes
