Design Notes

The design of EMX1 ngRNA includes the U6 promoter, spacer, guide RNA scaffold, and polyA tail.

Source

The original EMX1 ngRNA design containing spacer sequence was reported in Prime Editor 2 paper cited in References. With the help of TWIST Bioscience we synthesized that sequence and further pieced together with 65777 backbone in our lab.
